The two experiments we did recently with N2102Ep and NTERA were 50mg IP with 10ul of either nanobody or monoclonal antibody beads. Our results showed that anti-ORF1 monoclonal beads gave us much higher yield of ORF1p.

We got much better yield when we used ORF1 nanobody beads for MT302 IP. In that case, nanobody beads worked almost equally well compare to anti-ORF1 monoclonal beads (50mg IP with 5ul beads).

In a separate experiment where we did anti-ORF1 IP using both kinds of beads in MT302 and PA-1 cell lines. If you look at the Western result at the end, in PA-1 cells, monoclonal beads gave higher yield of ORF1p. In MT302, both beads gave similar yield of ORF1p, but the monoclonal anti-ORF1 pulldown more ORF2p at the same time. Scale: 50mg IP with 2.5, 5, and 10ul of LaORF1 beads or 10ul of anti-ORF1 beads.
