Media:

DMEM (High Glucose) (No sodium pyruvate) + 2mM Glutamine + 10% Foetal Bovine Serum (FBS):

• 500 ml media (https://www.thermofisher.com/order/catalog/product/11960044#/11960044)
• 50 ml FBS - F2442 sigma (https://www.sigmaaldrich.com/catalog/product/sigma/f2442?lang=en&region=US)
• 5 ml of Glutamine 100 mM (100X)
• 5 ml of pen/strep 10 000 mM (100X) – Final conc 100 U/ml

Cell passage: (75 cm2 plate)

• Aspire media from > 85% confluent plate.
• Add 3 mL DPBS (1X) and shake the plate.
• Aspire DPBS (1X).
• Add 2.5 TrypLE Express Enzyme (1X) in 37 degree for at least 3 mins.
• Neutralize with 2.5 mL of media.
