L1 is known to be active in pluripotent cells - somethings are known to be different about L1 RT activity in PC and DC (PA-1 as a model for pluripotent and report your data on these differences and/or point to references). We therefore sought to explore the interactomes of L1 in PA-1 PC and DC in an attempt to uncover information that may inform the mechanism of these differences.
IPs from PC by two antibodies (mono and nano) —> note differences, suggest different subpopulations are sampled as a result
IPs from DC by mono (compare PC to DC) —> note differences, suggest possible consequences of cell-type specific interactomes
Compare to HEK I-DIRT data, to CRC data (only proteins pass t-test 2x), and Vuong et al 2019 (because it is ES cells) —> note any similarities and differences to PC and DC states —> discuss massive differences between L1 interatcomes in nearly every dataset - point out biological and technical reasons —> Also, ZINC FINGERS!!!
Some cell biology follow-up of ~3-5 interesting hits (of which some ZNFs should be included).
So, in short we are reporting an exploratory study on L1 PA-1 interactomes that open the door for focussed studies and we introduce the concept of L1 interatcomes dynamics in different cell types - here controlled by looking at differentiation under a relatively uniform experimental framework compared to prior studies. Much of this will just be descriptive - it’s OK. Still we do need some additional validation to bring the paper into the realm of a good journal of e.g. impact ~15 I would say.
