^ First experiment (Ab concentration and lysate concentration test)
After I found the best conditions for Jess experiment I made a Jess to compare it with blot results from https://macromolecule-child.rockefeller.edu/#Oct%202020%20HEK%20293%20RRP6-3xFlag%20Flp-In%20TREX%20cell%20line%20DOX%20test%20induction
Conditions: 0,4 ug/ul is the best lysate concentration. 1:250 for anti-rrp6 Ab, 1:100 for anti-Flag Ab (info from Maria). All samples are the same as in WB (https://macromolecule-child.rockefeller.edu/#Oct%202020%20HEK%20293%20RRP6-3xFlag%20Flp-In%20TREX%20cell%20line%20DOX%20test%20induction)
separation time 35 min (max time). I asked Benjamine and decided to change separation time because the goal is to separate 100 and 103 kDa.

Comments from Nastya
This Jess was analyzed only by me. I folowed Benjamine protocols aboute analyse but it look like 35 min it's not enought to separate 100 and 103 kDa proteins from each other. 35 min it is a maximum setting that Jess could affort. After that experiment I didn't come back to this but another setting that you could adjust is separation voltage
