July 22, 2020 - CDI recombinant proteins

admin28th July 2020 at 2:05pm

Upon receiving, split each sample equally into 2 aliquots. One sent to

Kennedy C. Ukadike, MD, MS

Mustelin Lab, South Lake Union Campus

University of Washington School of Medicine

The other half, split into 5 aliquots and kept in the new -80C freezer in box "CDI recombinant proteins"

Also, run 250ng of each on a gel, followed by Sypro stain

Add LDS and 50mM DTT to each sample, heat @ 70C for 10’

Run 15-well Bis-Tris gel, Sypro stain; Loading order: • Unstained protein marker_1ul • Samples 1-13 • BSA_100ng (with 3ul of prestained marker in the same lane)

2-second exposure, auto tone

Sypro gel labeled with Molecular Mass of each protein

The recombinant proteins are human proteins expressed in yeast as a GST-tagged ectopic expression. So they will be ~26kDa heavier than the mass you find in Uniprot database.
