• I counted HEK293T_LD cells the standard was +/- trypan blue and came up with ~1.4M cells / ml - very few dead cells were observed in the culture.
  • Note: I used 1 part 0.4% w/v trypan blue to 5 parts cell culture (Luna proposes 1:1 using the same concentration)
  • cells were triturated 20 times using a p200 set to 200ul
• Using the Luna system I counts ranged from ~960k - 1.1M cells / ml depending on settings - with viability of ~99% seeming to be roughly accurate.​​
  • Settings giving 950k were: Dilution (1); Noise Reduction (4); Live Detection Sensitivity (5); Roundness (40); Min. Size (10); Max size (60)
  • it was observed that in some cases, double cells were counted as single cells [Image|
