Weigh out 50mg of each pre-mixed powder, add 200ul of extraction buffer
Extraction buffer: 20mM HEPES, pH7.4, 1% Tritonx-100, 500mM NaCl (with protease inhibitors), 1:4 (w/v)
5x 2sec @ 2 Amp with the new sonicator
Spin @ 20k rcf, 4°C for 10’ (Eppendorf Centrifuge 5417R)
Transfer supernatant to fresh tubes.
Determine the relative protein concentration of each batch using Bradford.
If all the samples have a similar concentration for the similar amount of mg, then we can proceed to check the %H (heavy labeling percentage)
