Cell Culture Room Etiquette

Before you start work This Checklist to be followed before beginning your work:

  1. Sign up on sheet
  2. Open hood, spritz liberally with 70% EtOH – this will evaporate while you do the next steps
  3. Place needed racks and pipettes into the hood - spritz all with 70% EtOH before placing, in order to avoid introducing contamination
  4. Take note of the state of the room - all in order?
  5. If not in order, take note of the last user and make sure to follow-up on un-tended issues
  6. Turn on centrifuge if needed at 4*C
  7. Check water level in 37*C water bath if it will be needed (halfway up the yellow rack is sufficient) - may take time to reach 37*C
  8. Do you have 70% EtOH?
  9. All the flasks, pipettes, media, etc. you will need?
  10. Place your items in the hood - everything entering the hood should be spritz with 70% EtOH in order to avoid introducing
  11. contamination
  12. Organize your work space - a well organize space reduces contamination - you don't want to reach over anything!


Before you leave the room This Checklist to be followed before leaving the room:

  1. Clean everything out of the hood (leave the Kimwipes, the open wipes live in there)
  2. Wash 70% EtOH or 10% bleach through the vacuum aspirator if you used it
  3. Swipe the working surface with 70% EtOH
  4. Close sash and turn on UV light
  5. Check trash - dispose if full
  6. Check bench area behind opposite the hood - should be cleared and swiped clean
  7. Turn off the refrigerated centrifuge
  8. Check 37*C water bath - refill if needed to halfway up the yellow rack
  9. Check cell incubators
  10. Temperature is 37*C?
  11. Water jacket is good? (alarm will sound if it is low)
  12. Humidity trays inside are good? (should have at least 1L of water)
  13. CO2 tanks are good? (Once tank is at 300 PSI, replace with spare tank. Label old tank "empty" and tell Rosemary to order a new tank)
  14. Make sure pipettes are fully stocked
  15. Makes sure anything you used will be replaced (e.g. Pasteur pipettes that need to be filled and autoclaved; boxes of sterile filter tips, etc.)
  16. Wash any bottles and cylinders, etc. (and remember to come back to collect and put away things after they have dried).
  17. Generally make sure the room is TIP FUCKING TOP.

Important: Make sure that you initial and date ANY media and reagents you use and keep.
